The Iowa team will face battle against the South Carolina Gamecocks in the national semifinals.
This is the second time in school history that Iowa has made it to the Final Four, with the previous being in 1993.
The Hawkeyes led for most of the game, but Kentucky tied the score at 68-68 in the fourth quarter before Iowa pulled away in the final minutes.
Due to COVID-19 regulations, there were few fans present at the game, which was played at the Alamodome in San Antonio, Texas.
The win over Kentucky was the Hawkeyes' seventh consecutive victory in the NCAA tournament.
Clark, a sophomore, is considered one of the best players in the country and is a finalist for the Naismith Trophy, which is awarded to the top women's college basketball player.
The championship game will be contested on April 5, following the Final Four games on April 1 and 3.
